Tal Danino: Programming bacteria to detect cancer (and maybe treat it)

01:03
Now, what you can't see
is that our genetic program

O que non poden ver é que
o noso programa xenético
01:06
instructs these bacteria
to each produce small molecules,
19
54285

ensina a estas bacterias a produciren
cada unha pequenas moléculas,
01:09
and these molecules travel between
the thousands of individual bacteria
20

as miles de bacterias individuais
01:13
telling them when to turn on and off.
21

01:15
And the bacteria synchronize
quite well at this scale,
22
63897
3251
E as bacterias sincronízanse
bastante ben a esta escala,
01:19
but because the molecule that synchronizes
them together can only travel so fast,
23
67148
3994
mais debido a que a molécula que as sincroniza
só pode viaxar a unha determinada velocidade,
01:23
in larger colonies of bacteria,
this results in traveling waves
24
71142
3576
en colonias maiores de bacterias,
isto resulta en ondas viaxando
01:26
between bacteria that are
far away from each other,
25
74718
2902
entre bacterias que están
lonxe unhas de outras,
01:29
and you can see these waves going
from right to left across the screen.

de dereita a esquerda na pantalla.
01:33
Now, our genetic program
relies on a natural phenomenon
27
81356
2788
O noso programa xenético
depende dun fenómeno natural,
01:36
called quorum sensing,
28
84144
1173
coñecido como a percepción do quórum,
01:37
in which bacteria trigger coordinated
and sometimes virulent behaviors
29
85317

no cal as bacterias causan comportamentos
cooperativos e virulentos
01:40
once they reach a critical density.
30

01:43
You can observe quorum sensing
in action in this movie,
31

Vexan a percepción do quórum
en acción nesta película,
01:46
where a growing colony of bacteria
only begins to glow

bacteria só brilla
01:49
once it reaches a high

Our genetic program

O noso programa xenético
01:54
continues producing these
rhythmic patterns of fluorescent proteins
35
102100

01:57
as the colony grows outwards.

This particular movie and experiment
we call The Supernova,

02:03
because it looks like an exploding star.
38
111310

02:06
Now, besides programming
these beautiful patterns,
39
114340
2390
Ademáis de programar
estes fermosos patróns,
02:08
I wondered, what else can we get
these bacteria to do?
40
116730
2591
Pregúntome, que máis podemos
obter destas bacterias?
02:11
And I decided to explore
how we can program bacteria
41
119321
2579
E decidín explorar como podemos
programar as bacterias
02:13
to detect and treat diseases
in our bodies like cancer.
42
121900
2789
para detectarmos e tratarmos
doenzas coma o cancro.
02:18
One of the surprising facts about bacteria
43
126009
2298
Un dos feitos máis
sorprendentes das bacterias
02:20
is that they can naturally grow
inside of tumors.
44
128307
3437
é que poden crecer de maneira
natural dentro de tumores.
02:23
This happens because typically tumors
45
131744
1764
Isto ocorre porque os tumores
02:25
are areas where the immune system
has no access,
46
133508
2415
son áreas onde o sistema
inmunitario non ten acceso
02:27
and so bacteria find these tumors
47
135923
1997
e así as bacterias atopan estes tumores
02:29
and use them as a safe haven
to grow and thrive.
48
137920
3831
e os usan como lugar protexido
para crecer e multiplicarse.
02:33
We started using probiotic bacteria
49
141751
1834
Comezamos usando bacterias probióticas
02:35
which are safe bacteria
that have a health benefit,
50
143585
2624
que son bacterias inócuas
con beneficios saudables,
02:38
and found that
when orally delivered to mice,
51
146209
2461
e descubrín que, ao seren
inxeridas oralmente por ratos,
02:40
these probiotics would selectively
grow inside of liver tumors.
52
148670
4156
estas probióticas crecían selectivamente
dentro de tumores de fígado.
02:44
We realized that the most convenient way
53
152826
1954
Decatámonos de que o xeito máis conveniente
02:46
to highlight the presence
of the probiotics,
54
154780
2081
de salientar a presenza das probióticas,
02:48
and hence, the presence of the tumors,
55
156861
1863
e, por tanto, a presenza dos tumores,
02:50
was to get these bacteria
to produce a signal
56
158724
2229
era facermos que as bacterias
emitisen un sinal
02:52
that would be detectable in the urine,
57
160953
2116
que sería detectable na urina,
02:55
and so we specifically
programmed these probiotics
58
163069
2412
e así programamos
específicamente estas probióticas
02:57
to make a molecule that would change
the color of your urine
59
165481
2879
para facermos unha molécula que
cambiase a cor da urina
03:00
to indicate the presence of cancer.
60
168360
2810
para indicar a presenza de cancro.
03:03
We went on to show that this technology
61
171170
2010
Demostramos que esta tecnoloxía
03:05
could sensitively and specifically
detect liver cancer,
62
173180
3190
podería detectar o cancro de fígado
de forma sensible e específica,
03:08
one that is challenging
to detect otherwise.
63
176370
2690
un cancro que é difícil
de atopar doutro xeito.
03:11
Now, since these bacteria
specifically localize to tumors,
64
179891
2999
Pois ben, xa que estas bacterias
se adhiren a tumores,
03:14
we've been programming them
to not only detect cancer
65
182890
2588
estamos a programalas
non só para detectarmos o cancro
03:17
but also to treat cancer
66
185478
1788
senón tamén para tratarmos o cancro
03:19
by producing therapeutic molecules
from within the tumor environment
67
187266
3227
producindo moléculas terapéuticas
dende o interior do tumor
03:22
that shrink the existing tumors,
68
190493
2322
que reducen os tumores existentes,
03:24
and we've been doing this
using quorum sensing programs
69
192815
2755
e fixémolo con programas
que usan a percepción do quórum
03:27
like you saw in the previous movies.
70
195570
1843
como viron en anteriores filmes.
03:29
Altogether, imagine in the future
taking a programmed probiotic
71

3697
Imaxinen inserirmos no futuro
unha probiótica programada
03:33
that could detect and treat cancer,
72

que poida detectar e tratar o cancro,
03:35
or even other diseases.
