ABOUT THE SPEAKER
Craig Venter - Biologist, genetics pioneer
In 2001, Craig Venter made headlines for sequencing the human genome. In 2003, he started mapping the ocean's biodiversity. And now he's created the first synthetic lifeforms -- microorganisms that can produce alternative fuels.

Why you should listen

Craig Venter, the man who led the private effort to sequence the human genome, is hard at work now on even more potentially world-changing projects.

First, there's his mission aboard the Sorcerer II, a 92-foot yacht, which, in 2006, finished its voyage around the globe to sample, catalouge and decode the genes of the ocean's unknown microorganisms. Quite a task, when you consider that there are tens of millions of microbes in a single drop of sea water. Then there's the J. Craig Venter Institute, a nonprofit dedicated to researching genomics and exploring its societal implications.

In 2005, Venter founded Synthetic Genomics, a private company with a provocative mission: to engineer new life forms. Its goal is to design, synthesize and assemble synthetic microorganisms that will produce alternative fuels, such as ethanol or hydrogen. He was on Time magzine's 2007 list of the 100 Most Influential People in the World.

In early 2008, scientists at the J. Craig Venter Institute announced that they had manufactured the entire genome of a bacterium by painstakingly stitching together its chemical components. By sequencing a genome, scientists can begin to custom-design bootable organisms, creating biological robots that can produce from scratch chemicals humans can use, such as biofuel. And in 2010, they announced, they had created "synthetic life" -- DNA created digitally, inserted into a living bacterium, and remaining alive.

You know, I've talked about some of these projects項目 before --

2000
00:21
about the human人的 genome基因組 and what that might威力 mean,
1
3000
4000
00:25
and discovering發現 new sets of genes基因.

3000
00:28
We're actually其實 starting開始 at a new point:
3
10000
3000
00:31
we've我們已經 been digitizing數字化 biology生物學,
4
13000
4000
00:35
and now we're trying to go from that digital數字 code
5
17000
3000
00:38
into a new phase of biology生物學
6
20000
2000
00:40
with designing設計 and synthesizing合成 life.
7
22000
3000
00:43
So, we've我們已經 always been trying to ask big questions問題.
8
25000
3000
00:48
"What is life?" is something that I think many許多 biologists生物學家
9
30000
2000
00:50
have been trying to understand理解
10
32000
2000
00:52
at various各個 levels水平.
11
34000
2000
00:57
paring削皮 it down to minimal最小 components組件.
13
39000
3000
01:01
We've我們已經 been digitizing數字化 it now for almost幾乎 20 years年份;
14
43000
2000
01:03
when we sequenced測序 the human人的 genome基因組,
15
45000
2000
01:05
it was going from the analog類似物 world世界 of biology生物學
16
47000
3000
01:08
into the digital數字 world世界 of the computer電腦.
17
50000
4000
01:12
Now we're trying to ask, "Can we regenerate再生 life
18
54000
4000
01:16
or can we create創建 new life
19
58000
2000
01:18
out of this digital數字 universe宇宙?"
20
60000
3000
01:21
This is the map地圖 of a small organism生物,

01:32
and we've我們已經 been trying to just see if
25
74000
2000
01:34
we can come up with an even smaller genome基因組.
26
76000
3000
01:38
We're able能夠 to knock out on the order訂購 of 100 genes基因
27
80000
2000
01:40
out of the 500 or so that are here.
28
82000
3000
01:43
When we look at its metabolic新陳代謝 map地圖,
29
85000
2000
01:45
it's relatively相對 simple簡單
30
87000
2000
01:49
trust相信 me, this is simple簡單 --
32
91000
2000
01:51
but when we look at all the genes基因
33
93000
2000
01:53
that we can knock out one at a time,
34
95000
3000
01:56
it's very unlikely不會 that this would yield產量
35
98000
2000
01:58
a living活的 cell細胞.
36
100000
2000
02:01
So we decided決定 the only way forward前鋒
37
103000
2000
02:03
was to actually其實 synthesize合成 this chromosome染色體
38
105000
3000
02:06
so we could vary變化 the components組件
39
108000
3000
02:09
to ask some of these most fundamental基本的 questions問題.
40
111000
4000
02:13
And so we started開始 down the road of:
41
115000
2000
02:15
can we synthesize合成 a chromosome染色體?
42
117000
3000
“我們能人工合成染色體嗎?”
02:19
Can chemistry化學 permit許可證 making製造
43
121000
2000
化學方法真的可以讓我們製造
02:21
these really large molecules分子
44
123000
2000
02:23
where we've我們已經 never been before?
45
125000
2000
02:25
And if we do, can we boot up a chromosome
46
127000
3000
02:28
A chromosome染色體, by the way, is just a piece of inert惰性的 chemical化學 material材料.
47
130000
3000
02:32
So, our pace步伐 of digitizing數字化 life has been increasing增加
48
134000
3000
02:35
at an exponential指數 pace步伐.
49
137000
3000
02:38
Our ability能力 to write the genetic遺傳 code
50
140000
3000
02:41
has been moving移動 pretty漂亮 slowly慢慢地
51
143000
2000
02:43
but has been increasing增加,
52
145000
3000
02:46
and our latest最新 point would put it on, now, an exponential指數 curve曲線.
53
148000
4000
02:51
We started開始 this over 15 years年份 ago.
54
153000
2000
02:53
It took several一些 stages階段, in fact事實,
55
155000
3000
02:56
starting開始 with a bioethical生命倫理學 review評論 before we did the first experiments實驗.
56
158000
3000
03:00
But it turns out synthesizing合成 DNA脫氧核糖核酸
57
162000
2000
03:02
is very difficult.
58
164000
2000
03:04
There are tens of thousands數千 of machines around the world世界
59
166000
3000
03:07
that make small pieces of DNA脫氧核糖核酸 --
60
169000
2000
03:09
30 to 50 letters in length
61
171000
3000
03:12
and it's a degenerate退化 process處理, so the longer you make the piece,
62
174000
3000
03:15
the more errors錯誤 there are.
63
177000
2000
03:17
So we had to create創建 a new method方法
64
179000
2000
03:19
for putting these little pieces together一起 and correct正確 all the errors錯誤.
65
181000
3000
03:23
And this was our first attempt嘗試, starting開始 with the digital數字 information信息
66
185000
3000
03:26
of the genome基因組 of phi X174.
67
188000
2000
03:28
It's a small virus病毒 that kills殺死 bacteria.
68
190000
3000
03:32
We designed設計 the pieces, went through通過 our error錯誤 correction更正
69
194000
3000
03:35
and had a DNA脫氧核糖核酸 molecule分子
70
197000
2000
03:37
of about 5,000 letters.
71
199000
3000
03:40
The exciting扣人心弦 phase came來了 when we took this piece of inert惰性的 chemical化學
72
202000
4000
03:44
and put it in the bacteria,
73
206000
2000
03:46
and the bacteria started開始 to read this genetic遺傳 code,
74
208000
4000
03:50
made製作 the viral病毒 particles粒子.
75
212000
2000
03:52
The viral病毒 particles粒子 then were released發布 from the cells細胞
76
214000
2000
03:54
and came來了 back and killed殺害 the E. coli大腸桿菌.
77
216000
3000
03:57
I was talking to the oil industry行業 recently最近
78
219000
3000
04:00
and I said they clearly明確地 understood了解 that model模型.
79
222000
3000
04:03
(Laughter
80
225000
3000
04:06
They laughed笑了 more than you guys are. (Laughter笑聲)
81
228000
3000
04:10
And so, we think this is a situation情況
82
232000
2000
04:12
where the software軟件 can actually其實 build建立 its own擁有 hardware硬件
83
234000
3000
04:15
in a biological生物 system系統.
84
237000
2000
04:17
But we wanted to go much larger:
85
239000
2000
04:19
we wanted to build建立 the entire整個 bacterial細菌 chromosome染色體 --
86
241000
3000
04:22
it's over 580,000 letters of genetic遺傳 code --
87
244000
4000
04:26
so we thought we'd星期三 build建立 them in cassettes盒子 the size尺寸 of the viruses病毒
88
248000
3000
04:29
so we could actually其實 vary變化 the cassettes盒子
89
251000
2000
04:31
to understand理解
90
253000
2000
來理解
04:33
what the actual實際 components組件 of a living活的 cell細胞 are.
91
255000
3000
04:36
Design設計 is critical危急,
92
258000
2000
04:38
and if you're starting開始 with digital數字 information信息 in the computer電腦,
93
260000
3000
04:41
that digital數字 information信息 has to be really accurate準確.
94
263000
4000
04:45
When we first sequenced測序 this genome基因組 in 1995,
95
267000
3000
04:48
the standard標準 of accuracy準確性 was one error錯誤 per 10,000 base基礎 pairs.
96
270000
4000
04:52
We actually其實 found發現, on resequencing測序 it,
97
274000
2000
04:54
30 errors錯誤; had we used that original原版的 sequence序列,
98
276000
3000
04:57
it never would have been able能夠 to be booted啟動 up.
99
279000
3000
05:00
Part部分 of the design設計 is designing設計 pieces
100
282000
2000
05:02
that are 50 letters long
101
284000
3000
05:05
that have to overlap交疊 with all the other 50-letter-信 pieces
102
287000
3000
05:08
to build建立 smaller subunits亞基
103
290000
2000
05:10
we have to design設計 so they can go together一起.
104
292000
3000
05:13
We design設計 unique獨特 elements分子 into this.
105
295000
3000
05:16
You may可能 have read that we put watermarks水印 in.
106
298000
2000
05:18
Think of this:
107
300000
2000
想想看
05:20
we have a four-letter四個字母 genetic遺傳 code -- A, C, G and T.
108
302000
3000
05:23
Triplets三胞胎 of those letters
109
305000
3000
05:26
code for roughly大致 20 amino氨基 acids,
110
308000
2000
05:28
such這樣 that there's a single letter designation指定
111
310000
3000
05:31
for each of the amino氨基 acids.
112
313000
2000
05:33
So we can use the genetic遺傳 code to write out words,
113
315000
3000
05:39
Initially原來, all we did was autograph簽名 it.
115
321000
2000
05:41
Some people were disappointed失望 there was not poetry詩歌.
116
323000
3000
05:44
We designed設計 these pieces so
117
326000
2000
05:46
we can just chew back with enzymes;
118
328000
3000
05:50
there are enzymes that repair修理 them and put them together一起.
119
332000
3000
05:53
And we started開始 making製造 pieces,
120
335000
2000
05:55
starting開始 with pieces that were 5,000 to 7,000 letters,
121
337000
4000
05:59
put those together一起 to make 24,000-letter-信 pieces,
122
341000
4000
06:03
then put sets of those going up to 72,000.
123
345000
4000
06:07
At each stage階段, we grew成長 up these pieces in abundance豐富
124
349000
2000
06:09
so we could sequence序列 them
125
351000
2000
06:11
because we're trying to create創建 a process處理 that's extremely非常 robust強大的
126
353000
3000
06:14
that you can see in a minute分鐘.
127
356000
3000
06:17
We're trying to get to the point of automation
128
359000
3000
06:20
So, this looks容貌 like a basketball籃球 playoff挑撥.
129
362000
2000
06:22
When we get into these really large pieces
130
364000
2000
06:24
over 100,000 base基礎 pairs,
131
366000
4000
06:28
they won't慣於 any longer grow增長 readily容易 in E. coli大腸桿菌 --
132
370000
2000
06:30
it exhausts尾氣 all the modern現代 tools工具 of molecular分子 biology生物學 --
133
372000
4000
06:34
and so we turned轉身 to other mechanisms機制.
134
376000
4000

08:14
Obviously明顯, with a virus病毒 it's pretty漂亮 simple簡單;

08:22
into eukaryotes真核生物 like ourselves我們自己:
171

08:24
you can just pop流行的 out the nucleus

08:26
and pop流行的 in another另一個 one,

08:31
With bacteria and Archaea, the chromosome染色體 is integrated集成 into the cell細胞,
175
493000

08:35
but we recently最近 showed顯示 that we can do a complete完成 transplant移植

roughly大致, these two are as distant遙遠 as human人的 and mice老鼠 --
180
510000
3000
08:51
we added添加 a few少數 extra額外 genes基因
181
513000
2000
08:53
so we could select選擇 for this chromosome染色體,
182

08:55
we digested消化 it with enzymes

and it was pretty漂亮 stunning令人驚嘆 when we put this in the cell細胞 --
185
521000
3000
09:02
and you'll你會 appreciate欣賞

09:04
our very sophisticated複雜的 graphics圖像 here.
187
526000
3000
09:07
The new chromosome染色體 went into the cell細胞.
188
529000
3000
09:10
In fact事實, we thought this might威力 be as far as it went,
189
532000
2000
09:12
but we tried試著 to design設計 the process處理 a little bit further進一步.
190
534000
3000
09:15
This is a major重大的 mechanism機制 of evolution演化 right here.
191
537000
3000
09:18
We find all kinds of species種類
192
540000
2000
09:20
that have taken採取 up a second第二 chromosome染色體
193
542000

09:28
So, people who think of evolution演化
197
550000
2000
09:30
as just one gene基因 changing改變 at a time
198
552000
2000
09:32
have missed錯過 much of biology生物學.
199
554000
3000
09:35
There are enzymes called restriction限制 enzymes
200
557000
2000
09:39
The chromosome染色體 that was in the cell細胞
202
561000
2000
09:41
doesn't have one;
203
563000
2000
09:43
the chromosome染色體 we put in does.
204
565000
2000
09:45
It got expressed表達 and it recognized認可
205
567000
2000
09:47
the other chromosome染色體 as foreign國外 material材料,
206
569000
3000
09:50
chewed咀嚼 it up, and so we ended結束 up
207
572000
2000
09:52
just with a cell細胞 with the new chromosome染色體.
208
574000
4000
09:56
It turned轉身 blue藍色 because of the genes基因 we put in it.
209
578000
3000
09:59
And with a very short period of time,
210
581000
2000
10:01
all the characteristics特點 of one species種類 were lost丟失
211
583000
3000
10:04
and it converted轉換 totally完全 into the new species種類
212
586000
3000
10:07
based基於 on the new software軟件 that we put in the cell細胞.
213
589000
3000
10:10
All the proteins蛋白質 changed,
214
592000
2000
10:12
the membranes changed;
215
594000
2000
10:14
when we read the genetic遺傳 code, it's exactly究竟 what we had transferred轉入 in.
216
596000
4000
10:18
So, this may可能 sound聲音 like genomic基因組 alchemy煉金術,
217
600000
3000
10:21
but we can, by moving移動 the software軟件 of DNA脫氧核糖核酸 around,
218
603000
4000
10:29
Now I've argued爭論, this is not genesis創世紀;
220
611000
2000
10:31
this is building建造 on three and a half billion十億 years年份 of evolution演化.
221
613000
4000
10:36
And I've argued爭論 that we're about to perhaps也許
222
618000
2000
10:38
create創建 a new version of the Cambrian寒武紀的 explosion爆炸,
223
620000
3000

10:49
I think this is pretty漂亮 obvious明顯 in terms條款 of some of the needs需求.
